So at the the weekend 6 of us biked from Momument to Brighton. Three of us did it single speed, I did it on my track bike, fixed! The ride was brilliant - pretty much back roads all the way when you get out of London - we followed this route: www.freewebs.com/cycleroutes/
There are a couple of really steep ascents - pretty treacherous on a fixed gear bike!
If you are thinking about doing it - JUST DO IT!
